Can samples be safely handled within a standard BSL-1 laboratory? Biosafety level 1 (BSL-1) applies to laboratory settings in which personnel work with low-risk microbes that pose little to no threat of infection in healthy adults.

Sample Quality

We emphasize the importance of good quality samples. For our Discovery-seq service, the following recommendations apply:

Cell viability of control samples is >70%.

Wells are seeded at a density of 2,500 to 10,000 cells per well (384-well plate).

Cells are washed in PBS.

Maximum volume left behind is 15 uL.

Plates are sealed with the original seal and parafilm around the borders.

Plates are frozen by covering them with dry ice for 10 minutes before transferring to -80°C.

Plates are labeled with same names provided in each sample submission sheet (1 sheet per plate).

Plates are shipped on dry ice.

I confirm that the plates have been properly sealed with the original plate seals and parafilm around the edges, and stabilized well within the package to avoid damage.

I understand that plates will be disposed after 6 months of storage at Single Cell Discoveries, if not processed before that time, counted from the date of arrival.

FAQ

Which types of samples are compatible with Discovery-seq?

For now we support cell lines of any animal species. We are also developing Discovery-seq for tissue samples and organoids.

How many cells do I need to provide per sample?

The ideal seeding density is between 5,000 and 10,000 cells, but we can work with at least 2,500 cells.

How should I prepare the cells?

We require the cells to be washed in PBS to avoid the interference with our lysis and reverse transcription.

What sort of analysis can you provide?

Our standard service includes the mapping of the sequencing reads, together with its quality metrics. We also provide an overview of the performance of each well in a plate, based on unique genes and UMI recovered.
