Can samples be safely handled within a standard BSL-1 laboratory? Biosafety level 1 (BSL-1) applies to laboratory settings in which personnel work with low-risk microbes that pose little to no threat of infection in healthy adults.

Sample Quality

Before our lab team starts processing samples with the 10x Genomics protocol, a careful quality check (QC) of the cell suspension(s) is performed. Please be aware that samples that do not meet QC requirements will NOT be processed.

Samples fail QC when:

The viability is lower than 70% (N/A to MeOH or formaldehyde fixed samples)

The multiplets/clumps, or debris in suspension is over 30% (by approximation, N/A to formaldehyde fixed samples)

Not enough cells are present

Viability is lower than 50% prior to fixation

Not enough cells/nuclei are present

If your samples don't reach the minimum viability requirement, please send an email to our lab team for more information.

FAQ

How many cells do I need to send per sample?

Ideally, we require a cell suspension with at least 500,000 cells per sample. If you are unable to achieve that, please contact us to discuss the possibilities.

Can I enrich my sample with FACS sorting?

Yes, before you send us your samples, you can perform FACS sorting to enrich your cell type of interest. Please bear in mind that we need at least 500,000 cells to start the 10x Genomics project.

How many cells should I aim for per sample?

Target cell numbers depend on your biological question and sample type. Typically, our customers target between 3,000 and 8,000 cells per sample. Schedule a call with one of our specialists to discuss your specific project.

How many reads per cell should I aim for?

The amount of reads depends on your biological question. Typically, we recommend a sequencing depth between 30,000 and 70,000 reads per cell for 10x Genomics projects. Schedule a call with one of our specialists to discuss your options.

How should I send you the samples?

We accept cryopreserved and methanol-fixed samples for the 3’ Single Cell Gene Expression services.
For the 5’ Single Cell Immune Profiling service, we accept cryopreserved cells.
If you are located close to our lab in Utrecht, the Netherlands, then we can discuss receiving a fresh sample as well.
