Can samples be safely handled within a standard BSL-1 laboratory? Biosafety level 1 (BSL-1) applies to laboratory settings in which personnel work with low-risk microbes that pose little to no threat of infection in healthy adults.

Sample Quality

We emphasize the importance of good quality samples. For our bulk RNA sequencing services, the following recommendations apply:

Tissues and/or cells were collected using a clean work area and clean tools (free of RNAses). If possible, surfaces were treated with RNAseZAP.

The process from cell isolation, cell sorting, or tissue dissection to storage in TRIzol (or similar buffer) was as short as possible, to prevent RNA degradation.

Tissue pieces in TRIzol were fully dissociated before storage.

For TRIzol samples, at least 90% of the total sample volume consists of TRIzol (10:1), to ensure protection of RNA integrity.

For TRIzol LS, at least 75% of the total sample volume consists of TRIzol (3:1), to ensure protection of RNA integrity.

TRIzol LS reagent was used when the starting material contains high levels of RNAse, such as spleen or pancreas samples.

TRIzol LS reagent should NOT be used undiluted with solid samples.

For low input samples, the cell number between samples is as equal as possible.

FAQ

How deep should I sequence my samples?

Sequencing depth depends on the number of samples, your budget, and your biological question. We offer two packages for bulk RNA sequencing: STANDARD, with 10 million sequencing reads per sample, and PLUS, with 20 million sequencing reads per sample.

Should I include technical or biological duplicates in my project?

Even bulk RNA sequencing is not without noise. Hence it is usually wise for downstream analysis purposes to consider triplicates or, at least, duplicates in your experimental set-up. We always recommend you include at least a second vial for each of the samples to ensure technical backup.
